The valine residue is highly conserved and there is a small physicochemical difference between valine and isoleucine. This variant is present in population databases (rs202068193, ExAC 0.01%). This missense change has been observed in individual(s) with hemochromatosis (PMID: 21411349). ClinVar contains an entry for this variant (Variation ID: 461194).
