not specified Uncertain:1
The c.2665T>C (p.F889L) alteration is located in exon 18 (coding exon 17) of the CTNNA3 gene. This alteration results from a T to C substitution at nucleotide position 2665, causing the phenylalanine (F) at amino acid position 889 to be replaced by a leucine (L). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Arrhythmogenic right ventricular dysplasia 13 Uncertain:1
This sequence change replaces phenylalanine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 889 of the CTNNA3 protein (p.Phe889Leu). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CTNNA3-related conditions. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CTNNA3 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
