Variant summary
Our verdict is Uncertain significance. The variant received 1 ACMG points: 3P and 2B. PM2PP2BP4_Moderate
The NM_003061.3(SLIT1):c.4351G>C(p.Glu1451Gln) variant causes a missense change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a benign outcome for this variant. 16/22 in silico tools predict a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. E1451K) has been classified as Uncertain significance.

The c.4351G>C (p.E1451Q) alteration is located in exon 36 (coding exon 36) of the SLIT1 gene. This alteration results from a G to C substitution at nucleotide position 4351, causing the glutamic acid (E) at amino acid position 1451 to be replaced by a glutamine (Q). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
