This sequence change replaces glycine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 50 of the ISCU protein (p.Gly50Glu). This variant is present in population databases (rs267607190, gnomAD 0.003%). This missense change has been observed in individual(s) with myopathy with lactic acidosis (PMID: 19567699). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 783). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change affects ISCU function (PMID: 24573684). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively.
