The encoded protein is part of a large protein complex called ASCOM, which has been shown to be a transcriptional regulator of the beta-globin and estrogen receptor genes. This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 5464 of the KMT2D protein (p.Thr5464Met). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with Kabuki syndrome (PMID: 20711175, 25142838). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 7539).
