This protein is cytoplasmic but is often anchored to a peroxisomal membrane where it forms a heteromeric complex and plays a role in the import of proteins into peroxisomes and peroxisome biogenesis. Mutations in this gene have been associated with complementation group 1 peroxisomal disorders such as neonatal adrenoleukodystrophy, infantile Refsum disease, and Zellweger syndrome. Multiple computational tools predict a significant impact on normal splicing: Three predict the variant abolishes a canonical 5' splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 8e-06 in 251308 control chromosomes. c.2926+2T>C has been reported in the literature in individuals affected with Zellweger Syndrome (Walter_2001, Cheema_2020, Rosewich_2005). These data indicate that the variant is associated with disease. One publication reports experimental evidence evaluating an impact on protein expression, finding no expression of PEX1 protein in cells that are compound heterozygous for this variant and c.2097dup (Walter_2001). One clinical diagnostic laboratory has submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation, classifying the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
