This missense change has been observed in individual(s) with Hermansky-Pudlak syndrome and/or ocular albinism (PMID: 12442288, 29345414). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is also known as c.922T>C. ClinVar contains an entry for this variant (Variation ID: 1452567). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt HPS1 protein function with a positive predictive value of 80%. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.716T>C has been reported in the literature in multiple individuals affected with Hermansky-Pudlak Syndrome or Inherited albinism (example, Lasseaux_2017, Hermos_2002). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
