This sequence change replaces tyrosine, which is neutral and polar, with serine, which is neutral and polar, at codon 154 of the SERPING1 protein (p.Tyr154Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with angioedema (PMID: 23123409; Invitae). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SERPING1 protein function with a positive predictive value of 80%. This variant disrupts the p.Tyr154 amino acid residue in SERPING1. Other variant(s) that disrupt this residue have been observed in individuals with SERPING1-related conditions (PMID: 21864911, 23123409; Invitae), which suggests that this may be a clinically significant amino acid residue. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively.
