The p.R24S variant (also known as c.72G>T), located in coding exon 1 of the RAD51D gene, results from a G to T substitution at nucleotide position 72. The arginine at codon 24 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

This sequence change replaces arginine, which is basic and polar, with serine, which is neutral and polar, at codon 24 of the RAD51D protein (p.Arg24Ser). This variant is present in population databases (rs28363257, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with RAD51D-related conditions. ClinVar contains an entry for this variant (Variation ID: 239401).
