This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 435 of the PRF1 protein (p.Thr435Met). This variant is present in population databases (rs28933376, gnomAD 0.009%). This missense change has been observed in individual(s) with hemophagocytic lymphohistiocytosis (PMID: 12599189, 25233452, 32542393). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 13717).
