Variant summary: HFE c.277G>C (p.Gly93Arg) results in a non-conservative amino acid change located in the MHC class I alpha chain, alpha1 alpha2 domains (IPR001039) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251474 control chromosomes. c.277G>C has been reported in the literature in trans with p.Cys282Tyr (a common low-penetrance pathogenic variant) in at least 2 related individuals affected with Hemochromatosis Type 1 (example, Barton_1999). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 10575540). ClinVar contains an entry for this variant (Variation ID: 13). Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic. -
