Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251392 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1694G>A has been reported in the literature in an individual affected with Familial Hypercholesterolemia (Sanchez-Hernandez _2016). This report however does not provide unequivocal conclusions about association of the variant with Familial Hypercholesterolemia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Different variants affecting the same codon (p.G565A and p.G565V) have been reported in patients in HGMD database and ClinVar, cited as pathogenic/likely pathogenic. A ClinVar submitter (evaluation after 2014) cites the variant as likely pathogenic based on evidence of presence of the variant in 1 compound heterozygote FH index case and co-segregation in 1 family member.
