A knockout mouse model has been used to study the effect of mutations in this gene. The mouse gene is also located on the X chromosome, however, unlike the human gene it is not subject to X inactivation. Mutations in this gene are associated with oral-facial-digital syndrome type I and Simpson-Golabi-Behmel syndrome type 2. This variant has been observed in individual(s) with oral-facial-digital syndrome type 1 (OFD1) (PMID: 18546297; Invitae). This variant is not present in population databases (gnomAD no frequency). This variant, c.1219_1221del, results in the deletion of 1 amino acid(s) of the OFD1 protein (p.Glu407del), but otherwise preserves the integrity of the reading frame. This variant also falls at the last nucleotide of exon 12, which is part of the consensus splice site for this exon. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively.
