The ALPL c.1540G>A; p.Ala514Thr variant (rs367657406) is reported in the literature in an individual affected with infantile hypophosphatasia that also carried a known pathogenic ALPL variant (Tenorio 2017). The p.Ala514Thr variant is reported in ClinVar (Variation ID: 554421) and is found in the general population with an overall allele frequency of 0.009% (24/265430 alleles) in the Genome Aggregation Database. The alanine at codon 514 is weakly conserved, and computational analyses (SIFT, PolyPhen-2) predict that this variant is tolerated.
