This protein plays a role in the sonic hedgehog (SHH) signaling pathway through the regulation of GLI transcription factors. It functions as a negative regulator of the SHH pathway by preventing inappropriate activation of GLI2 in the absence of ligand, and as a positive regulator by preventing the processing of GLI3 into its repressor form. The c.2425C>T (p.R809W) alteration is located in exon 12 (coding exon 11) of the KIF7 gene. This alteration results from a C to T substitution at nucleotide position 2425, causing the arginine (R) at amino acid position 809 to be replaced by a tryptophan (W). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This variant is present in population databases (rs367734857, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with KIF7-related conditions.
