The c.2405-4G>A intronic variant results from a G to A substitution 4 nucleotides upstream from coding exon 16 in the FLNA gene. This nucleotide position is poorly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing. Based on data from gnomAD, the A allele has an overall frequency of <0.01% (3/179449) total alleles studied, with 0 hemizygote(s) observed. The highest observed frequency was <0.01% (1/12329) of African alleles. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
