MYBPC3 is expressed exclusively in heart muscle and is a key regulator of cardiac contraction. Mutations in this gene are a frequent cause of familial hypertrophic cardiomyopathy. The p.N251I variant (also known as c.752A>T), located in coding exon 6 of the MYBPC3 gene, results from an A to T substitution at nucleotide position 752. The asparagine at codon 251 is replaced by isoleucine, an amino acid with dissimilar properties. This alteration was detected in an individual from the Jackson Heart Study cohort (Bick AG. Am J Hum Genet. 2012;91(3):513-9). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
