Genes affected
MMP13 (HGNC:7159): (matrix metallopeptidase 13) This gene encodes a member of the peptidase M10 family of matrix metalloproteinases (MMPs). Proteins in this family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. The encoded preproprotein is proteolytically processed to generate the mature protease. This protease cleaves type II collagen more efficiently than types I and III. It may be involved in articular cartilage turnover and cartilage pathophysiology associated with osteoarthritis. Mutations in this gene are associated with metaphyseal anadysplasia. This gene is part of a cluster of MMP genes on chromosome 11. [provided by RefSeq, Jan 2016]

Pathogenic, criteria provided, single submitterclinical testingInvitaeAug 31, 2022For these reasons, this variant has been classified as Pathogenic. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. ClinVar contains an entry for this variant (Variation ID: 183688). This premature translational stop signal has been observed in individual(s) with autosomal recessive metaphyseal anadysplasia (PMID: 24781753). It has also been observed to segregate with disease in related individuals. This variant is present in population databases (rs369083541, gnomAD 0.004%). This sequence change creates a premature translational stop signal (p.Arg109*) in the MMP13 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in MMP13 are known to be pathogenic (PMID: 24781753, 31413057). -
