MYO15A (HGNC:7594): (myosin XVA) This gene encodes an unconventional myosin. This protein differs from other myosins in that it has a long N-terminal extension preceding the conserved motor domain. Studies in mice suggest that this protein is necessary for actin organization in the hair cells of the cochlea. Mutations in this gene have been associated with profound, congenital, neurosensory, nonsyndromal deafness. This gene is located within the Smith-Magenis syndrome region on chromosome 17. Read-through transcripts containing an upstream gene and this gene have been identified, but they are not thought to encode a fusion protein. Several alternatively spliced transcript variants have been described, but their full length sequences have not been determined. [provided by RefSeq, Jul 2008]

The Arg1794Cys variant in MYO15A has not been previously reported in individuals with hearing loss, but has been identified in 0.01% (1/8406) of European Americ an chromosomes by the NHLBI Exome Sequencing Project (http://evs.gs.washington.e du/EVS/). Although this variant has been seen in the general population, its fre quency is not high enough to rule out a pathogenic role. Computational predictio n tools and conservation analyses suggest that the Arg1794Cys variant may impact the protein, though this information is not predictive enough to determine path ogenicity. In summary, the clinical significance of the Arg1794Cys variant is un certain. -

The c.5380C>T (p.R1794C) alteration is located in exon 21 (coding exon 20) of the MYO15A gene. This alteration results from a C to T substitution at nucleotide position 5380, causing the arginine (R) at amino acid position 1794 to be replaced by a cysteine (C). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
