Valine (Val) at position 9435 is not well conserved in evolution and pand a carries an isoleucine (Ile) at this position, raising the possibility that thi s change may be tolerated. Additional computational analyses (biochemical amino acid properties, AlignGVGD, and PolyPhen2) also suggest that the Val9435Ile vari ant may not impact the protein, though this information is not predictive enough to rule out pathogenicity. In summary, although the available data raise the po ssibility that the Val9435Ile variant may be benign, additional studies are need ed to fully assess its clinical significance. - |

This frequency is not significantly higher than estimated for a pathogenic variant in TTN causing Dilated Cardiomyopathy (4.9e-05 vs 0.00039), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.28303G>A in individuals affected with Dilated Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported.
