The p.Val14357Ala variant in TTN has been identified by our laboratory in 1 adul t with HCM and AFib; however, this individual also carried a pathogenic MYH7 var iant. The p.Val14357Ala variant has also been identified in 4/66492 European chr omosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.o rg; dbSNP rs370067597). Computational prediction tools and conservation analysis do not provide strong support for or against an impact to the protein. In summa ry, the clinical significance of the p.Val14357Ala variant is uncertain. -
Variant summary: TTN c.43070T>C (p.Val14357Ala), also reported as c.50774T>C p.Val16925Ala, results in a non-conservative amino acid change located in the I-band of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.6e-05 in 247360 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.43070T>C has been reported in the literature in at least 1 individual affected with dilated cardiomyopathy (example, Mazzarotto_2020). These report(s) do not provide unequivocal conclusions about association of the variant with TTN-related conditions. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
