The c.1044G>T (p.E348D) alteration is located in exon 10 (coding exon 8) of the MEGF10 gene. This alteration results from a G to T substitution at nucleotide position 1044, causing the glutamic acid (E) at amino acid position 348 to be replaced by an aspartic acid (D). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

This sequence change replaces glutamic acid with aspartic acid at codon 348 of the MEGF10 protein (p.Glu348Asp). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and aspartic acid. This variant is present in population databases (rs370219469, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with MEGF10-related conditions. ClinVar contains an entry for this variant (Variation ID: 472732). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0".
