Variant summary: PCDH15 c.1006C>T (p.Arg336X) results in a premature termination codon, predicted to cause a truncation of the encoded protein or absence of the protein due to nonsense mediated decay, which are commonly known mechanisms for disease. The variant was absent in 251302 control chromosomes (gnomAD). c.1006C>T has been reported in the literature in individuals affected with Usher Syndrome Type 1F (e.g., Bonnet_2016). These data suggest the variant is very likely to be associated with disease. The following publication was ascertained in the context of this evaluation (PMID: 27460420). Three submitters have reported clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as pathogenic/likely pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -
