Desmogleins are calcium-binding transmembrane glycoprotein components of desmosomes, cell-cell junctions between epithelial, myocardial, and other cell types. The encoded preproprotein is proteolytically processed to generate the mature glycoprotein. This gene is present in a gene cluster with other desmoglein gene family members on chromosome 18. Mutations in this gene have been associated with arrhythmogenic right ventricular dysplasia, familial, 10. [provided by RefSeq, Jan 2016]

The methionine at codon 875 is replaced by valine, an amino acid with highly similar properties. This variant co-occurred with a PKP2 nonsense mutation in an individual from an arrhythmogenic right ventricular cardiomyopathy cohort (Cox MG et al. Circulation, 2011 Jun;123:2690-700). This variant has also been detected in a dilated cardiomyopathy (DCM) cohort and a cohort referred for DCM genetic testing; however, clinical details were limited and other genetic findings were present (Herkert JC et al. Genet Med, 2018 11;20:1374-1386; (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
