The arginine at codon 1119 is replaced by histidine, an amino acid with highly similar properties. This variant was detected in a cohort of 63 patients clinically diagnosed with Coffin-Siris syndrome and classified as a variant of unknown significance by the authors (Santen GW et al. Hum Mutat, 2013 Nov;34:1519-28). This variant was also detected in a cohort of 11 patients with autism and mild-severe intellectual disability and dysmorphic features who underwent whole exome sequencing (Vandeweyer G et al. Am J Med Genet C Semin Med Genet, 2014 Sep;166C:315-26). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
