This was consistent with a diagnosis of tyrosinemia type 1 in this patient, his sister was also previously diagnosed with tyrosinemia type 1 (Dou et al., 2013). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The W152X variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). We interpret W152X as a pathogenic variant. -
