This sequence change replaces threonine, which is neutral and polar, with methionine, which is neutral and non-polar, at codon 42 of the TRDN protein (p.Thr42Met). This variant is present in population databases (rs371627659, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with TRDN-related conditions. ClinVar contains an entry for this variant (Variation ID: 229348). The p.T42M variant (also known as c.125C>T), located in coding exon 2 of the TRDN gene, results from a C to T substitution at nucleotide position 125. The threonine at codon 42 is replaced by methionine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species; however, methionine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
