The encoded protein mediates facilitated bidirectional glucose transport. Because of its low affinity for glucose, it has been suggested as a glucose sensor. Mutations in this gene are associated with susceptibility to diseases, including Fanconi-Bickel syndrome and noninsulin-dependent diabetes mellitus (NIDDM). This sequence change affects a donor splice site in intron 7 of the SLC2A2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in SLC2A2 are known to be pathogenic (PMID: 11810292). This variant is present in population databases (rs371977235, gnomAD 0.002%). Disruption of this splice site has been observed in individuals with Fanconi-Bickel syndrome (PMID: 11810292). ClinVar contains an entry for this variant (Variation ID: 444008). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
