This variant was observed in the ICSL laboratory as part of a predisposition screen in an ostensibly healthy population. In the published literature, this variant has been reported in an individual with personal history of a carotid paraganglioma (PMID: 28384794 (2017)). The frequency of this variant in the general population, 0.000012 (3/251388 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is damaging. Based on the available information, we are unable to determine the clinical significance of this variant. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.V393M variant (also known as c.1177G>A), located in coding exon 9 of the SDHA gene, results from a G to A substitution at nucleotide position 1177. The valine at codon 393 is replaced by methionine, an amino acid with highly similar properties. This alteration has been reported in an individual diagnosed with a carotid paraganglioma at age 63 who had no family history of paraganglioma or pheochromocytoma (Bausch B et al. JAMA Oncol, 2017 Sep;3:1204-1212). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
