This variant is present in population databases (rs373407069, gnomAD 0.006%). This missense change has been observed in individual(s) with male breast cancer (PMID: 30613976). ClinVar contains an entry for this variant (Variation ID: 140973). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt RAD50 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
The p.R1198Q variant (also known as c.3593G>A), located in coding exon 23 of the RAD50 gene, results from a G to A substitution at nucleotide position 3593. The arginine at codon 1198 is replaced by glutamine, an amino acid with highly similar properties. In one study, this alteration was identified in a neuroblastoma patient (Lasorsa VA et al. Oncotarget, 2016 Apr;7:21840-52). In another study, this alteration was identified in a male breast cancer patient (Rizzolo P et al. Int J Cancer, 2019 07;145:390-400). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
