The previously defined group FANCH is the same as FANCA. Fanconi anemia is a genetically heterogeneous recessive disorder characterized by cytogenetic instability, hypersensitivity to DNA crosslinking agents, increased chromosomal breakage, and defective DNA repair. The members of the Fanconi anemia complementation group do not share sequence similarity; they are related by their assembly into a common nuclear protein complex. This sequence change has been described in the gnomAD database with a frequency of 0.007% in the European subpopulation (dbSNP rs3736772). The p.Pro1812Thr change affects a poorly conserved amino acid residue located in a domain of the FANCM protein that is not known to be functional. The p.Pro1812Thr substitution appears to be benign using several in-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL). Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.Pro1812Thr change remains unknown at this time -
