Myosin-binding protein C is a myosin-associated protein found in the cross-bridge-bearing zone (C region) of A bands in striated muscle. MYBPC3 is expressed exclusively in heart muscle and is a key regulator of cardiac contraction. Mutations in this gene are a frequent cause of familial hypertrophic cardiomyopathy. This variant results from a C to T substitution at nucleotide position 3123. This nucleotide substitution does not change the glycine at codon 1041. A minigene study indicated that this variant may not have a significant impact on splicing (Ito K et al. Proc Natl Acad Sci U S A, 2017 Jul;114:7689-7694). This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will result in the creation or strengthening of a novel splice donor site. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
