This sequence change replaces alanine with threonine at codon 71 of the SMPD1 protein (p.Ala71Thr). The alanine residue is weakly conserved and there is a small physicochemical difference between alanine and threonine. This variant is present in population databases (rs375224040, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with SMPD1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
