The encoded protein associates with the sperm annulus, as part of the septin complex, through direct interaction with septin 4, during sperm terminal differentiation. This sequence change replaces arginine, which is basic and polar, with serine, which is neutral and polar, at codon 25 of the DNAJB13 protein (p.Arg25Ser). This variant is present in population databases (rs375603870, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with DNAJB13-related conditions. ClinVar contains an entry for this variant (Variation ID: 1442004). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
