The p.S188N variant (also known as c.563G>A), located in coding exon 1 of the RSPH4A gene, results from a G to A substitution at nucleotide position 563. The serine at codon 188 is replaced by asparagine, an amino acid with highly similar properties. This variant was previously reported in the SNPDatabase as rs376372993. Based on data from the NHLBI Exome Sequencing Project (ESP), the A allele has an overall frequency of approximately 0.01% (1/13006) total alleles studied and 0.01% (1/8600) European American alleles. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear. -
Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This sequence change replaces serine with asparagine at codon 188 of the RSPH4A protein (p.Ser188Asn). The serine residue is moderately conserved and there is a small physicochemical difference between serine and asparagine. This variant is present in population databases (rs376372993, ExAC 0.01%). This variant has not been reported in the literature in individuals with RSPH4A-related disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
