The p.Arg2057His va riant in TRIOBP has not been previously reported in individuals with hearing los s, but has been identified in 5/62222 of European chromosomes by the Exome Aggre gation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs377068258). Alt hough this variant has been seen in the general population, its frequency is not high enough to rule out a pathogenic role. Computational prediction tools and c onservation analyses suggest that the p.Arg2057His variant may not impact the pr otein, and two mammals (opossum and platypus) have a histidine (His) at this pos ition suggesting that this variant may be tolerated.
