The p.M1146L variant (also known as c.3436A>T), located in coding exon 26 of the SBF2 gene, results from an A to T substitution at nucleotide position 3436. The methionine at codon 1146 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Charcot-Marie-Tooth disease type 4 Uncertain:1
This sequence change replaces methionine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 1146 of the SBF2 protein (p.Met1146Leu). This variant is present in population databases (rs377277274, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with SBF2-related conditions. ClinVar contains an entry for this variant (Variation ID: 543338).
