Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 0.00071 in 1586860 control chromosomes, predominantly at a frequency of 0.00084 within the Non-Finnish European subpopulation in the gnomAD database, including 2 homozygotes. This frequency is close the maximum estimated for a pathogenic variant in MYBPC3 suggesting a benign role. c.926+8C>T has been reported in the literature in individuals affected with Hypertrophic Cardiomyopathy. These reports do not provide unequivocal conclusions about association of the variant with Hypertrophic Cardiomyopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 23782526). Eight submitters have cited clinical-significance assessments for this variant to ClinVar after 2014 and classified as benign/likely benign (n=7) and VUS (n=1).
