Members of this family recognize the GATA motif which is present in the promoters of many genes. Mutations in this gene have been associated with cardiac septal defects. Additionally, alterations in gene expression have been associated with several cancer types. The aspartic acid at codon 210 is replaced by asparagine, an amino acid with highly similar properties. This variant was reported in an individual with unspecified congenital heart disease (Wang E et al. PLoS ONE. 2013;8(4):e62138). This variant was previously reported in the SNPDatabase as rs377673676. Based on data from the NHLBI Exome Sequencing Project (ESP), the A allele has an overall frequency of approximately 0.01% (1/13006) total alleles studied, having been observed in 0.02% (1/4406) African American alleles. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
