These data indicate that the variant is associated with disease. Four clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. The arginine at codon 361 is replaced by histidine, an amino acid with highly similar properties. This mutation has been reported in multiple individuals with a clinical diagnosis of juvenile polyposis syndrome (JPS) and several also with a diagnosis of hereditary hemorrhagic telangiectasia (HHT) (Kim IJ et al.
