The c.730_731delAG pathogenic mutation, located in coding exon 7 of the APC gene, results from a deletion of two nucleotides at nucleotide positions 730 to 731, causing a translational frameshift with a predicted alternate stop codon (p.R244Vfs*7). This variant has been observed in individuals with a personal and/or family history that is consistent with APC-associated polyposis conditions (García-Lozano JR et al. Genet. Test., 2005;9:37-40; Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this variant is interpreted as a disease-causing mutation. -
