This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 183 of the TRPV4 protein (p.Glu183Lys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with clinical features of TRPV4-related conditions (PMID: 20503319; Invitae). ClinVar contains an entry for this variant (Variation ID: 18437). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt TRPV4 protein function with a positive predictive value of 95%. Experimental studies have shown that this missense change affects TRPV4 function (PMID: 22702953). This variant disrupts the p.Gly183 amino acid residue in TRPV4. Other variant(s) that disrupt this residue have been observed in individuals with TRPV4-related conditions (PMID: 20503319, 25802885; Invitae), which suggests that this may be a clinically significant amino acid residue. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively.
