This sequence change replaces leucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 185 of the ETHE1 protein (p.Leu185Arg). This variant is present in population databases (rs387906987, gnomAD 0.004%). This missense change has been observed in individual(s) with ethylmalonic encephalopathy (PMID: 14732903, 19289697, 20528888, 27830356). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 30725).
