The encoded protein plays a role in cell adhesion, motility and proliferation, and is a critical mediator of apoptotic cell phagocytosis as well as amyloid-beta peptide uptake in the brain. Expression of this gene may be associated with schizophrenia, and mutations in this gene are a cause of early-onset myopathy, areflexia, respiratory distress, and dysphagia (EMARDD) as well as congenital myopathy with minicores. This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 71 of the MEGF10 protein (p.Arg71Trp). This variant is present in population databases (rs387907074, gnomAD 0.006%). This missense change has been observed in individual(s) with clinical features of early-onset myopathy, areflexia, respiratory distress and dysphasia (EMARDD) (PMID: 22371254). ClinVar contains an entry for this variant (Variation ID: 30967). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
