Genes affected
STK4 (HGNC:11408): (serine/threonine kinase 4) The protein encoded by this gene is a cytoplasmic kinase that is structurally similar to the yeast Ste20p kinase, which acts upstream of the stress-induced mitogen-activated protein kinase cascade. The encoded protein can phosphorylate myelin basic protein and undergoes autophosphorylation. A caspase-cleaved fragment of the encoded protein has been shown to be capable of phosphorylating histone H2B. The particular phosphorylation catalyzed by this protein has been correlated with apoptosis, and it's possible that this protein induces the chromatin condensation observed in this process. STK4-related disorder Pathogenic:1
Pathogenic, criteria provided, single submitter | clinical testing | PreventionGenetics, part of Exact Sciences | Aug 28, 2023 | The STK4 c.349C>T variant is predicted to result in premature protein termination (p.Arg117*). This variant in the homozygous condition was reported in individuals with primary immunodeficiency (see example: Table S1, Thaventhiran. 2020. PubMed ID: 32499645; Table S2, Turro. 2020. PubMed ID: 32581362). This variant is reported in 0.0040% of alleles in individuals of European (Finnish) descent in gnomAD (http://gnomad.broadinstitute.org/variant/20-43610573-C-T). Nonsense variants in STK4 are expected to be pathogenic. This variant is interpreted as pathogenic. - |
