Missense variant in a coldspot region where missense variants are very unlikely to be pathogenic (PMID:31911673). -
The p.P920T variant (also known as c.2758C>A), located in coding exon 10 of the BRCA2 gene, results from a C to A substitution at nucleotide position 2758. The proline at codon 920 is replaced by threonine, an amino acid with highly similar properties. This alteration was identified in a proband of a family with four cases of breast cancer from a cohort of 350 Swiss breast and ovarian cancer families (Maillet P et al, Cancer Genet. Cytogenet. 2006 Aug; 169(1):62-8). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces proline with threonine at codon 920 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been performed for this variant. This variant has been detected in a breast cancer case-control meta-analysis in 1/60466 cases and 1/53461 unaffected individuals (PMID: 33471991; Leiden Open Variation Database DB-ID BRCA2_002553). This variant also has been reported in an individual affected with breast cancer (PMID:16875939). A multifactorial analysis has reported a likelihood ratio for pathogenicity based on personal and family history of 0.521 from log(LR)=-0.282874525 for 1 carrier (PMID: 31853058). This variant has been identified in 1/250768 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
