This variant is expected to result in an absent or non-functional protein product. The c.5754_5755delTA pathogenic mutation, located in coding exon 10 of the BRCA2 gene, results from a deletion of two nucleotides at nucleotide positions 5754 to 5755, causing a translational frameshift with a predicted alternate stop codon (p.H1918Qfs*5). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). The p.His1918fs variant in BRCA2 has been reported in 1 European individual with breast cancer (Hansen 2011). This variant has also been identified in 1/66606 of European chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs397507803).
