The p.His1918fs variant in BRCA2 has been reported in 1 European individual with breast cancer (Hansen 2011). This variant has also been identified in 1/66606 of European chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs397507803). This variant is predicted to cause a frameshift, which alters the protein’s amino acid sequence beginning at position 1918 and leads to a premature termination codon 5 amino acids downstream. This alteration is then predicted to lead to a truncated or absent protein. Heterozygous loss of BRCA2 function an established disease mechanism in BRCA2-associated cancers. In summary, the p.His1918fs variant meets our criteria to be classified as pathogenic for BRCA2-associated cancers in autosomal dominant manner (http://www.partners.org/personalizedmedicince/LMM) based upon its predicted impact to the protein and low frequency in controls. -

This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
