Submissions by phenotype
not provided Pathogenic:1
This variant disrupts a canonical splice-acceptor site and interferes with normal BRCA2 mRNA splicing. The variant has not been reported in individuals with BRCA2-related diseases in the published literature. It also has not been reported in large, multi-ethnic general populations (http://gnomad.broadinstitute.org). Based on the available information, this variant is classified as pathogenic. -
Hereditary breast ovarian cancer syndrome Pathogenic:1
This sequence change affects an acceptor splice site in intron 14 of the BRCA2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with breast and/or ovarian cancer (PMID: 24156927, 30287823). ClinVar contains an entry for this variant (Variation ID: 1400967). Studies have shown that disruption of this splice site alters mRNA splicing and is expected to lead to the loss of protein expression (PMID: 22505045, 31191615). For these reasons, this variant has been classified as Pathogenic. -
