This sequence change creates a premature translational stop signal (p.Leu151Glyfs*133) in the KCNQ1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in KCNQ1 are known to be pathogenic (PMID: 9323054, 19862833). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with autosomal recessive Jervell and Lange-Nielsen syndrome (PMID: 10077519). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 53045). For these reasons, this variant has been classified as Pathogenic. -
Cardiovascular phenotype Pathogenic:1
The c.451_452delCT pathogenic mutation, located in coding exon 2 of the KCNQ1 gene, results from a deletion of two nucleotides at nucleotide positions 451 to 452, causing a translational frameshift with a predicted alternate stop codon (p.L151Gfs*133). This variant has been identified in the homozygous state in two siblings of Amish descent with features consistent with Jervell and Lange-Nielsen syndrome (Chen Q et al. Circulation, 1999 Mar;99:1344-7). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
