The variant allele was found at a frequency of 1.2e-05 in 251248 control chromosomes (gnomAD). c.1006_1007insG has been reported in the literature in individuals affected with cystic fibrosis or chronic pancreatitis (examples: Ooi_2012, Beauchamp_2019). The following publications have been ascertained in the context of this evaluation (PMID: 31036917, 22658665). Six submitters(including CFTR2 expert panel) have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. -

The c.1006_1007insG pathogenic mutation, located in coding exon 8 of the CFTR gene, results from an insertion of one nucleotide at position 1006, causing a translational frameshift with a predicted alternate stop codon (p.I336Sfs*28). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
